It has long been recognized that many proteins most likely start to fold cotranslationally, but it is only in recent years that experimental and computational methods to study cotranslational folding have become available. We have developed a method in which so-called translational arrest peptides are used as force sensor to detect cotranslational folding reactions. The methodology – Force Profile Analysis – makes it possible to follow folding reactions for both soluble and membrane proteins with up-to single-residue resolution.
