Abstract:
ICP0 is an E3 ubiquitin ligase expressed by herpes simplex virus-1 (HSV-1) which mediates the proteasomal degradation of several host proteins that are important in antiviral responses such as PML and IFI16. We have recently discovered a complex of two tegument proteins that can also cause the degradation of many of the same host targets as ICP0, but via a distinct mechanism that relies on protein kinase activity. Loss of function of both this complex and ICP0 is required to attenuate HSV-1 replication, suggesting HSV-1 has evolved two independent mechanisms to target host factors involved in intrinsic and innate immune responses.
