Abstract:
We have been involved in collaborative studies, especially with the group of Gavin Screaton, to understand antibody responses to SARS-CoV-2 spike. Single cell sequencing has been used to isolate mAbs from convalescent patients. Together with sera from patients infected with all major variants to date these reagents have been used in biophysical analyses, virus and pseudovirus neutralisation studies and extensive structural analyses (X-ray and cryo-EM). In addition we have developed novel computational methods in an attempt to build an understanding of how the antibody responses are shaped by the antigen at the residue level and to help our thinking about how this will affect virus breakthrough. Some results are already published in a series of 2021 Cell papers, but I will also present recent work which extends the scope of results and the antigenic mapping methods.
