CSSB Seminar Series - FRANCESCA BOTTANELLI

Abstract:
In our laboratory, we integrate endogenous tagging with live-cell super-resolution STED microscopy to illuminate dynamic processes occurring at the nanoscale and in the unperturbed physiological environment of the cell. We have generated a comprehensive library of over 100 tagged proteins involved in trafficking. The ability to explore cellular dynamics at sub 50 nm resolution is revealing novel and unexplored sorting mechanisms. We have recently provided functional characterization of a novel sorting compartment that we named ARF1 compartment. ARF1 compartments orchestrate clathrin-dependent post-Golgi trafficking, challenging the conventional notion of long-range transport through small clathrin-coated vesicles. Our work suggests the presence of a tubulo-vesicular network linking the Golgi apparatus with the endosomal system and the plasma membrane.
