Abstract:
In the human large intestine, bacteria within the microbiota grow in a soup of complex dietary polysaccharides, but the outer membrane of Gram-negative bacteria presents a prodigious barrier to the uptake of such nutrients. Using cryoEM we have been studying the mechanism of carbohydrate uptake by a novel class of transporters. I will describe the structure of a novel 'utilisome' complex for the integrated processing and transport of complex polysaccharides, and describe the biochemical and image processing approaches used to deal with heterogeneity and conformational change in this system, yielding new biological insight.
